Abstract

The possibility of using nanocarriers based on single walled carbon nanotubes (SWCNTs) to deliver genetic material into E.coli cells, BY3-tobacco cells and chick embryo cells of five days old is discussed. Using single walled CNTs (SWCNTs) at the concentration of 0.3% of PL-PEG-5000-NH2-SWCNT the E.coli cells were genetically transformed with the plasmid construct of 300 ng pEGFP-C1 and a transient expression of the green fluorescent protein was shown in the E.coli cells. Using SWCNTs at the concentration of 5ml of PL-PEG-5000-NH2-SWCNT the BY3-tobacco cell protoplasts were genetically transformed with the plasmid construct (4μg/ml; 6μg/ml) of pEGFP-C1 along with 1X PBS (Phosphate Buffered Saline) and a transient expression of the green fluorescent protein was shown in the BY3-tobacco cell protoplasts . Using SWCNTs at the concentration of 10μl of PL-PEG-5000-NH2-SWNT the chick embryos were genetically transformed with the plasmid construct (300mg/10μl) of pEGFP-C1 and a transient expression of the green fluorescent protein was shown in the chick embryo cells. The PL-PEG-5000-NH2-SWCNT based nanocarriers demonstrated their applicability for the transformation of bacterial cells, BY3-tobacco cell protoplasts and chick embryo cells.
